چکیده

Human chromosomes from cultures of peripheral lymphocytes were prepared for electron microscopy by surface spreading and critical-point drying. Total and partial determinations of chromosomal dry mass were made from contrast in electron microscopic negatives. The distribution of dry weights was analyzed with respect to the Denver-Chicago classifications. There is considerable variability of mass within each group, but no statistically significant differences between four donors. A single chromosome can be correctly classified from its total mass and the ratio of long- to short-arm mass.

Analysis of human chromosomal variants by quantitative electron microscopy. I. Group D chromosome with giant satellites.

HE so-called “normal” variations in the human karyotype found in clinically Tnormal individuals have been useful for linkage studies as chromosome markers (e.g., No. 1 and No. 16 chromosomes) (DONAHUE et al. 1968; MAGENIS, HECHT and LOVIEN 1970). The old order Amish in the United States are an inbred group that has been extensively studied genetically ( MCKUSICK, HOSTETLER and EGELAND 1964). Du...

Internal ultrastructure of human chromosomes.

By preparing metaphase chromosome spreads on Aclar (polychlorotrifluoroethylene fluoropolymer) film and incorporating a conventional karyotyping system, it was possible to observe the internal ultrastructure of individual chromosomes with known karyotype by means of transmission electron microscopy. Essentially, all 46 chromosomes per human cell could be examined. Application of this method to ...

Karyotyping human and mouse cells using probes from single-sorted chromosomes and open source software.

Multispectral karyotyping analyzes all chromosomes in a single cell by labeling them with chromosome-specific probes conjugated to unique combinations of fluorophores. Currently available multispectral karyotyping systems require the purchase of specialized equipment and reagents. However, conventional laser scanning confocal microscopes that are capable of separating multiple overlapping emiss...

Phenotypes and Karyotypes of Human Malignant Mesothelioma Cell Lines

BACKGROUND Malignant mesothelioma is an aggressive tumour of serosal surfaces most commonly pleura. Characterised cell lines represent a valuable tool to study the biology of mesothelioma. The aim of this study was to develop and biologically characterise six malignant mesothelioma cell lines to evaluate their potential as models of human malignant mesothelioma. METHODS Five lines were initia...
